Genome Imaging Facility

The newly established Genome Imaging Facility (GIF) provides tools for cytogenetic analysis of the entire genome as well as specific probes targeting regions of interest. These tools include metaphase preparations from cultured cell lines and human peripheral blood along with FISH using locus specific probes (LSP) that are designed and labeled to meet experimental requirements. Spectral Karyotyping (SKY) and whole chromosome paint for human and mouse samples are also part of the service.

The core currently generates “in house” chromosome painting probes custom made using different combinations of heptans and fluorochromes dNTPs. Human and mouse single chromosome painting probes are generated from flow-sorted chromosomes and PCR-labeled through the incorporation of spectrally distinct fluorochromes. Hybridized chromosomes can be visualized using an epifluorescence microscope. We also generate LSP probes from BAC DNA for genes of interest. All labelings, hybridizations and analysis are performed in the core facility. Investigators will be provided with digital images of the analyzed cases as well as a summary of the abnormalities that are observed.
