Gene Targeting

The Gene Targeting Facility is a centralized facility for the production of gene-targeted mice. The services provided by the facility allow researchers to modify the mouse genome through methods of gene targeting or BAC transgenesis in embryonic stem (ES) cells and to introduce these changes into the mouse genome by the generation of targeted ES cells and chimeric mice. The facility also works in conjunction with the Gene Modification Facility to generate knockout mice.

  • Consultation on targeting vector construction, ES cell culture, ES cell screening methods, mouse genotyping and maintenance of gene targeted mice
  • Chimera production
    • Blastocyst injection
    • 6-8 cell embryro injection for high-yield chimera production
  • ES Cell Culture Service for 129Sv, B6 and 129V/B6 ES cell lines, including:
    • Construct electroporation
    • Isolation of targeted clones
    • Preparation for blastocyst injection
