Regulation of tumor metastasis by S100A4

This laboratory focuses on the regulation of tumor metastasis by S100A4, a calcium-regulated myosin binding protein expressed in both tumor cells and cells of the immune system. We have developed a floxed S100A4 mouse that allows us to ask how S100A4 expression in stromal cells contributes to tumorigenesis and metastasis. We have identified S100A4 as a critical regulator of macrophage invasion. S100A4 acts by regulating the dynamics and degradative functions of podosome rosettes, actin-rich structures that perform adhesive and invasive functions in macrophages. We are combining cell biological and animal-based studies to define how S100A4 regulates both the actin- and microtubule-based cytoskeletons in macrophages.

The postdoctoral project is directed to how S100A4-mediated regulation of macrophage function promotes a permissive metastatic environment. The successful applicant should have strong expertise in cell biology, biochemistry and mammalian cell culture.  Experience with mouse tumor models will be an asset.
